3. Synthesis, anticonvulsant and toxicity evaluation of 4-Amino-5-phenyl-4H[1,2,4]triazole-3-thiol derivatives

Abstract: A series of new 1,2,4-triazole derivatives were prepared by condensing the methyl benzoate and methyl salicylate with suitable reagents. All the titled compound structures were established on the basis of their elemental analysis, FTIR, 1H NMR and Mass Spectral data. All the titled compounds were screened for their anticonvulsant activity in the MES model and were compared with the standard drugs phenytoin sodium and carbamazepine. Out of the total titled compounds studied PS01, PS06, PS07, PS09 and PS10 showed comparable MES activity to phenytoin and carbamazepine after 0.5h. Compound PS09 showed to be more potent than carbamazepine after 4h. Compound PS01 and PS 08 showed lower neurotoxicity than phenytoin.
